چکیده

Background Sf-9 cells, derived from Spodoptera frugiperda, are widely used for recombinant protein production using the baculovirus expression vector system (BEVS). However, this results in a productive viral infection and cell lysis. Therefore, a non-lytic, plasmid-based expression system for suspension Sf-9 cells would be a valuable alternative to the BEVS for rapid, scalable, and highyielding recombinant protein production and for the generation of stable Sf-9 cell lines [4]. In this work, we present a simple, efficient and cost-effective plasmidbased method for transient expression of recombinant proteins in Sf-9 cells cultivated in serum-free suspension mode in a high-throughput culture system, TubeSpin bioreactor 50 tubes (TubeSpins).

Transient gene expression with CHO cells in conditioned medium: a study using TubeSpin® bioreactors

Background Transient gene expression (TGE) allows rapid protein production in mammalian cells and has become an important tool in the pharmaceutical product development pipeline [1]. Polyethylenimine (PEI)-mediated, high-density transfection allowed to express recombinant proteins at yields exceeding 1 g/L in only a few weeks [2]. Although highly efficient protocols are available, volumetric sc...

Baculovirus-directed expression of rabbit UDP-glucuronosyltransferases in Spodoptera frugiperda cells.

The rabbit liver UDP-glucuronosyltransferase (UGT) cDNAs that encode the 4-hydroxybiphenyl UGT2B13 and 4-nitrophenol UGT1A6 have been cloned into baculovirus. Spodoptera frugiperda (SF-9) cells infected with the UGT recombinant baculovirus produced significant amounts of protein, which was detected by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, immunoblot analysis, and pulse chas...

A Model to Study the Phenotypic Changes of Insect Cell Transfection by Copepod Super Green Fluorescent Protein (cop-GFP) in Baculovirus Expression System

Background: Baculovirus expression system is one of the most attractive and powerful eukaryotic expression systems for the production of recombinant proteins. The presence of a biomarker is required to monitor transfection efficiency or protein expression levels in insect cells. Methods: The aim of this study was to construct a baculovirus expression vector encoding a copepod super green fluore...

Cationic lipid-mediated co-transfection of insect cells

Current co-transfection protocols use calcium phosphate precipitation for cultured insect (Sf-9) cells followed by multiple agarose overlays to purify recombinant baculoviruses (1). We report a more effective and convenient method that uses the cationic lipid N[1,1-(2,3-dioleyloxy)propoyl]-N,N,N,trimethylammonium chloride, or DOTMA (2, 3), for cotransfections of Sf-9 cells and uses 96-well micr...

Rapid recombinant protein production from pools of transposon-generated CHO cells

Transient gene expression (TGE) is the most commonly used technology for the rapid production of moderate quantities of recombinant proteins for preclinical studies or for analytical assay development [1,2]. Whereas TGE can provide milligram to gram amounts of recombinant proteins within a time period as short as few days, technical limitations and high costs of plasmid DNA hamper the applicati...
